摘要
目的:观察增液承气汤对老年津亏便秘小鼠氧化应激的影响。方法:将42只小鼠随机分为空白组、便秘衰老组(简称便衰组)、增液承气汤组(简称增承组)、西沙必利组(简称西药组)。其中便衰组12只,其余组各10只。采用限水,叠加复方地芬诺酯灌胃,同时10%D-半乳糖皮下注射6周建立老年便秘模型。从第4周起分别给予相应药物干预3周。观察整体状态及小鼠体重变化并检测小肠墨汁推进率、大肠和皮肤含水率;血清、皮和肺超氧化物歧化酶(SOD),血清、皮肤组织羟脯氨酸(HYP)含量。结果 :从第4周开始给予相应处理后,西药组、增承组小鼠体重较便衰组小鼠有一定程度恢复(P<0.05,P<0.01),以第6周末最为明显,且增承组体重增加最为显著(P<0.01)。小鼠墨汁推进率、大肠含水率、皮肤含水率,血清SOD、HPY及皮肤SOD、HPY和肺SOD活性等,增承组与便衰组比较,差异均有非常显著性意义(P<0.01)。结论:增液承气汤能增强老年津亏便秘小鼠的抗氧化能力,具有抗氧化应激作用。
Objective: To observe the effects of Zengye Chengqi Decoction on oxidative stress in the aged mice model offluid-insufficiency constipation. Methods. Forty-two mice were randomized into blank control group, model group, Zengye Chengqi Decoction group and Cisapride group, and the mice were marked with picric acid. The model group had 12 mice, and the other groups had 10 in each group. Mice model of fluid-insufficiency constipation was induced by water limit, intragastric administration of diphenoxylate, and subcutaneous injection of 10% D-galactose for 6 weeks. From week 4, the mice were given the corresponding treatment for 3 weeks following the experimental design. During the experiment, the general health state and the change of body weight were monitored. After treatment, the small intestinal ink propelling ratio, and water content in the large intestine and skin were examined, and superoxide dismutase(SOD) activity in the serum, skin and lung as well as hydroxyproline(HYP) activity in the serum and skin was detected. Results. Compared with the model group, the body weight of mice in Zengye Chengqi Decoction group and Cisapride group was recovered to some degrees from week 4(P 〈 0.05, P〈 0.01), and the increase of body weight was most obvious in Zengye Chengqi Decoction group at the end of week 6(P 〈 0.01). The small intestinal ink propelling ratio, water content in the large intestine and skin, serum and skin SOD and HYP activities, and lung SOD activity were improved in ZengTe Chengqi Decoction group as compared with those in the model group (P 〈 0.01). Conclusion: Zengye Chengqi Decoction has anti-oxidative stress in aged mice with fluid-insufficiency constipation through enhancing SOD activity and inhibiting oxygen free radical.
