摘要
目的观察阿维A对HaCaT细胞表达水通道蛋白3(AQP3)mRNA的影响及增液汤含药血清的调节作用。方法应用不同浓度阿维A及增液汤干预HaCaT细胞,荧光定量反转录酶-聚合酶链反应(Reverse transcriptionpolymerase chain reaction,RT-PCR)测定HaCaT细胞AQP3 mRNA的表达。结果阿维A溶液干预下的HaCaT细胞AQP3 mRNA表达下降,呈剂量依赖性;增液汤含药血清干预下HaCaT细胞AQP3 mRNA的表达上升,呈剂量依赖性;HaCaT细胞在增液汤含药血清与阿维A共同干预下的AQP3 mRNA表达水平高于同等浓度阿维A溶液干预的表达。结论阿维A溶液下调HaCaT细胞AQP3 mRNA表达,增液汤含药血清可以抑制这种下调作用。
Objective In order to investigate the influence of acitretin solution on the expression of A QP3 mRNA in HaCaT cells and study the regulation of Zengyetang drug serum. Methods We used aeitretin solution and Zengyetang drug serum in different concentration to treat HaCaT cells and adopted RT-PCR method to detect A QP3 mRNA of HaCaT cells. Results The expression level of A QP3 mRNA in HaCaT cells treated with acitretin solution declined in a dose-dependent manner. The expression level ofA QP3 mRNA in HaCaT cells treated with Zengyetang drug serum raised and also showed dose-dependence. The expression level of A QP3 mRNA HaCaT cells treated with both Zengyetang drug serum and acitretin solution was higher than that only treated with the same concentration of acitretin. Conclusion The expression of A QP3 mRNA in HaCaT cells treated with acitretin solution declines. And this effect can be held up by Zengyetang drug serum.
