芜菁花叶病毒的分离鉴定及其多克隆抗体的制备与应用 被引量:3

摘要 从呈花叶症状的芥菜病株上分离到毒源,经CP基因克隆、序列分析以及电镜观察鉴定为芜菁花叶病毒(turnip mosaic virus,TuMV)。利用提纯的病毒免疫新西兰大白兔,获得TuMV多克隆抗体,其效价为1∶40 000,多抗仅对TuMV起特异性反应。以1∶10 000稀释度为TuMV多抗最适工作浓度,建立TuMV间接酶联免疫吸附测定法(ID-ELISA),该方法检测到提纯病毒的绝对量约1.765 ng。说明本试验所建立的TuMV ID-ELISA检测方法可有效运用于TuMV的检测。
