摘要 目的:筛选超声法提取贯叶连翘总黄酮的最佳工艺条件。方法:采用均匀设计法考察乙醇浓度、乙醇用量、提取时间、提取温度、超声提取次数等5个因素对贯叶连翘总黄酮提取的影响;利用紫外-可见分光光度法测定总黄酮的含量;以贯叶连翘总黄酮的得率为指标进行工艺条件的优化。结果:最佳提取工艺为乙醇浓度60%,乙醇用量20ml/g,每次超声提取时间43min,提取温度52℃,超声提取3次。结论:该提取工艺省时节能,可为工业化生产提供理论依据。 Objective: To optimize the extraction condition of total flavones from Hypericum perforatum by ultrasonic wave. Method: The uniform design method was applied to investigate the effects of alcohol content, the quantity of solvent, extraction time, extraction temperature, and the operation times on the result of extraction. The content of total flavone of Hypericum perforatum was determined by ultraviolet-visible spectrophotometry. The extraction conditions were optimized with the extraction yield of total Hypericum perforatum flavone as index. Result: The optimal extraction conditions were as follows: extracting with 60% alcohol, the quantity of solvent 20 ml/g, extraction time 43 minutes, temperature at 52 ℃, extract for 3 times. Conclusion: The extraction process is reasonable and feasible, and it provides theoretic basis for the industrialized production.
