贯叶连翘金丝桃素提取工艺的优化及含量的测定 被引量:4

摘要 以乙醇溶液为提取溶剂,采用加热回流法提取贯叶连翘(Hypreicum perforatum L.)金丝桃素,设计正交试验考察液料比、乙醇溶液体积分数、提取次数和提取时间对金丝桃素提取量的影响,优化提取工艺,并建立金丝桃素含量测定的紫外分光光度法。结果表明,采用紫外分光光度法测定金丝桃素含量,选择589 nm作为检测波长,精确度、稳定性、重复性良好,加样回收率合格。液料比对金丝桃素提取量有显著影响,优化的贯叶连翘金丝桃素提取工艺条件为液料比V乙醇∶m贯叶连翘=10∶1(mL/g)、乙醇溶液体积分数70%、提取时间1 h、提取3次,此条件下金丝桃素提取量为25.26 mg/g。 Using ethanol solution as solvent, hypericin was extracted from Hypreicum perforatum L. by heating reflux method. The effects of liquid to solid ratio, volume fraction of ethanol solution, extraction times and extraction time on the yield of hypericin was studied by orthogonal experiment to optimize the extraction technology. And ultraviolet spectrophotometry method for detecting the content of hypericin was established. The results showed that determining the content of hypericin by ultravi- olet spectrophotometry with 589 nm as the detection wavelength was accurate, stabile and repeatable, with eligible recovery rate. The optimum extraction process was Vethanol:msample =10:1 (mL/g), volume fraction of ethanol solution at 70%, extracting for 1 h and 3 times. The hypericin yield of H. perforatum was 25.26 mg/g under the optimum extraction process.
