摘要 目的建立肠缺血再灌注致急性呼吸窘迫综合征(ARDS)模型,观察巨噬细胞移动抑制因子(MIF)在血清和肺组织的表达,以探讨巨噬细胞移动抑制因子的表达在模型组中的意义。方法健康雄性SPF级SD大鼠40只,随机分为对照组(10只),肠缺血组(10只),肠缺血再灌注组(20只),肠缺血再灌注组筛选呼吸频率>28次/分且PaO2≤(60 mmHg),PaCO2<35 mmHg作为模型组。检测血清中MIF浓度变化,取肺组织用4%多聚甲醛固定24 h,石蜡包埋,制作5μm切片备用。免疫组化测定MIF在各组大鼠肺组织中的表达情况。结果模型组大鼠血清中MIF浓度明显升高(P<0.01),肺组织免疫组化表达明显增强(P<0.01)。结论肠缺血再灌注损伤致ARDS的发生几率较高,血清及肺组织中MIF表达增强可能是肠缺血再灌注损伤免疫炎症反应致多器官功能障碍造成ARDS的重要机制之一。 Objective Establishment of intestinal ischemia-reperfusion model of acute respiratory distress syndrome(ARDS),observation of expression of macrophage migration inhibitory factor(MIF) in serum and lung tissue and discussion of significance of MIF in the model group.Methods 40 healthy male SPF SD rats were randomly divided into control group(10 rats),intestinal ischemia group(10 rats) and intestinal ischemia-reperfusion group(20 rats).In the intestinal ischemia-reperfusion group,the rats with screening respiratory rate 28 beats / min,and PaO2 ≤(60 mmHg) PaCO2 35 mmHg were included.To examine the changes of MIF in serum,these lung tissue were fixed with 4% paraformaldehyde for 24 h,then embedded in paraffin,made to 5μm slices.MIF expression in lung tissue was measured by immunohistochemistry.Results MIF concentration in the serum of model group was significantly higher(P0.01),the immunohistochemical expression of lung tissue was significantly increased(P0.01).Conclusion Higher probability of occurrence of ARDS caused by intestinal ischemia-reperfusion injury is associated with increased expression of MIF in serum and lung tissue,it may be one of the important mechanisms of ARDS in multiple organ dysfunction caused by injury in immune and inflammatory response induced in intestinal ischemia-reperfusion.
