低分子肝素对电烧伤大鼠组织血管内皮生长因子与血小板内皮细胞黏附分子表达的影响 被引量:11

摘要 目的观察低分子肝素对电烧伤大鼠局部组织血管内皮生长因子及血小板内皮细胞黏附分子影响。方法 60只Wistar大鼠制成右后肢深度电烧伤模型,随机分为治疗组及空白对照组,治疗组30只大鼠自伤后当天起每日两次皮下注射低分子肝素;空白对照组伤后除不注射低分子肝素外,其余处理同治疗组。大体及组织学观察大鼠创面变化,取局部组织采用免疫组化法观察血管内皮生长因子和血小板内皮细胞黏附分子的表达。结果大体及组织学观察治疗组创面愈合较快,镜下见散在炎性细胞浸润,管腔内少量淤血,少许内皮细胞剥脱;对照组可见大量炎性细胞浸润,内皮细胞坏死脱落,血栓形成。治疗组血管内皮生长因子阳性表达明显高于对照组,呈逐渐增高的趋势,伤后10d两组差异具有统计学意义(P<0.01)。CD31阳性表达比较,伤后10d治疗组高于对照组(P<0.01)。两组局部组织中微血管密度值进行比较,可见治疗组均较对照组高,统计学分析显示,伤后第5、10天两者间的差异有统计学意义(P<0.05或P<0.01)。结论低分子肝素可增加局部受损组织血管内皮生长因子及血小板内皮细胞黏附分子的表达,促进了受损血管的恢复及新生血管的形成。 Objective To observe the effects of low molecular heparin on expression of CD31 and vascular endothelial growth factor(VEGF) in electric burned rats local tissue. Methods The tissue-injury model of high-voltage electrical burn on right hind limb of rats was established. Healthy Wistar rats were randomly divided into two groups (thirty in each group): low molecular weight heparin(LMWH)-theated group and control group. Subcutaneous LMWH was given to the therapy group of post-burn,twice everyday. Blank control group after injury was treated as the treatment group except injection of LMWH.To observe the wound morphology and histological change in the two groups, immunohistochemistry was used to examine the expression of VEGF and CD31. Results Gross and histological observation showed treatment group wound healed quickly, and scattered inflammatory cells infiltration,a small amount of gore in lumen and a little endothelial cell stripped were found under microscope. Control group showed inflammatory cells infiltration, endothelial cell necrosis falling off and thrombosis.The treatment group expressd higher positive VEGF than the control group, especially in 10 days than the control group(P0.01). CD31 positive expression, 10 days after the injury in the treatment group was significantly higher than the control group (P0.01). Local MVD value in the treatment group was higher than that of the control group, the difference between 5 and 10 days of the after injury statistically significant (P 0.05 or P 0.01).Conclusions Low molecular heparin can increase VECF and CD31 expression in local damaged tissues, promote the restoration of the damaged blood vessels and the formation of new blood vessels, which may be one of the effective mechanisms.
