摘要
目的:探讨ATRA作用于两种细胞系(3AO,ES_2)后两种粘附分子E-cad、CD44在mRNA水平和蛋白水平上的变化。方法:用RT-PCR检测3AO,ES_2两种细胞系经ATRA作用后E-cad,CD44在mRNA水平的变化,应用Western blot检测3AO,ES_2两种细胞系经ATRA作用后E-cad,CD44在蛋白水平上的变化。结果:两种细胞系经ATRA处理后,E-cad在mRNA水平及蛋白水平上表达均增加,统计学差异有显著性P<0.05;CD44在mRNA水平上表达减少,统计学显示差异有显著性,P<0.05;在蛋白水平上的表达有下降趋势,但未显示出明显的统计学差异,P>0.05。结论:ATTRA可能通过改变癌细胞表面的粘附分子来抑制卵巢癌细胞的侵袭和远处转移。
Objective: To observe the effect of all-trans retinoic acid (ATRA) on the expression of E-cadherin and CD44 in 3AO and ES2 ovarian carcinoma cell lines. Methods: E-cadherin and CD44 transcriptional levels were detected by RT-PCR and the protein levels were detected by Western blot. Results: ATRA suppressed the protein expression of CD44 and increased the expression of E-cadherin at the mRNA and protein levels in 3AO and FS2 cell lines. Conclusion: ATRA can change the expression of E-cadherin and CD44 in 3AO and ES2 cells, indicating an effective and novel approach for clinical treatment of invasion and implantation of human ovarian cancer.
