卵巢上皮性癌组织中E钙黏素基因启动子区甲基化及其表达 被引量:3

摘要 卵巢上皮性癌(卵巢癌)是一种常见的、高病死率的女性生殖系统恶性肿瘤,大多数患者死于肿瘤盆腹腔种植转移引起的并发症,多种黏附分子参与上述过程。E钙黏素(E—cadherin,E—cad)是钙依赖性上皮细胞黏附分子,以同种亲和的方式与邻近上皮细胞相互作用,其基因的失活会导致细胞间黏附力下降,癌细胞易于从原发灶脱落,进而向周围、远处组织浸润转移。研究表明,在卵巢癌组织中E-cad表达降低;临床分期越高,表达降低越明显;且其表达与生存时间有关。引起E—cad表达降低的机制主要有:基因突变、杂合性缺失、甲基化等。DNA甲基化作为转录水平的DNA修饰方式之一,在调节基因表达中起了重要的作用。本研究检测了卵巢癌组织中E.cad基因的启动子区5’胞嘧啶-磷酸-鸟嘌呤二核苷酸(cytosine phosphate guanosine,CpG)岛甲基化状况及其蛋白的表达,并结合临床病理特征进行分析,以探讨卵巢癌浸润转移的机制。
