摘要
目的建立去甲基左氧氟沙星手性分离的高效液相色谱方法。方法色谱柱采用十八烷基硅烷键和硅胶为填充剂,流动相为L-异亮氨酸硫酸铜溶液-甲醇(85:15),流速为1.0mL/min;检测波长330nm,柱温:40℃。结果在此条件下,去甲基左氧氟沙星及其对映体能够达到基线分离,分离度为2.7。结论简单快速,重现性好,能够用于去甲基左氧氟沙星对映体的分离和纯度考察。
Objective To establish a high performance liquid chromatographic method for chiral separation of the enantiomer from desmethyl levofloxacin and quantitative determination of bulk drugs. Methods Analysis was carried out on a C18 column with a L- isoleucine and copper sulfate solution-methol(85: 15) as mobile phase. The flow rate is about 1.0mL per minute and UV detection wavelength was 330nm, the column temperature was 40℃. Results A baseline separation of desmethyl levofloxacin enantiomers was achieved and the resolution was between the enantiomers under the above chromatographic conditions. Conclusion This method is simple, rapid and with good reproducibility and it can be used for separation and purity of desmethyl levofloxacin.
