摘要 目的探讨金花茶种子诱导人前列腺癌PC3细胞凋亡的机制。方法用不同浓度金花茶种子乙醇提取物的正丁醇萃取物(CCEB)干预人前列腺癌PC3细胞,分别采用流式细胞仪、Western印记法等检测其对细胞凋亡及Bax、Bcl-2等表达的影响。结果 CCEB作用于人前列腺癌PC3细胞后,引起PC3细胞DNA片段化率增大,线粒体膜电位下降,细胞内caspase-3活性增高,Bax蛋白表达增加,Bcl-2蛋白表达减少,胞浆中细胞色素c含量增加。结论 CCEB诱导PC3细胞凋亡与caspase依赖性线粒体途径相关。 Objective To investigate the potential mechanism of Camellia chrysantha (Hu) Tuyama seeds - induced apoptosis in PC3 human prostate cancer cells. Methods PC3 human prostate cancer cells were treated with the solution of BuOH fraction of the EtOH extract from the seeds of Camellia chrysantha (Hu) Tuyama (CCEB) at different concentrations. The cell cycle distribu- tions and apoptosis were detected by flow cytometry ( FCM ) , apoptosis associated proteins ( Bax, Bcl - 2 ) were determined by western blot. Results DNA fragmentation was increased, mitochondrial membrane potential was decreased and the activity of caspase - 3 was enhanced in the cells treated with CCEB. The activation of Bax and deactivation of Bcl - 2 were observed after the treatment. The release of cytochrome c from mitochondria to cytosol was enhanced by CCEB. Conclusion These results indicate that CCEB can induce apoptosis in PC3 human prostate cancer cells by the mitoehondria - mediated caspase - dependent pathway.
