摘要
目的:观察当归不同组分对人前列腺癌PC3细胞、人宫颈癌HeLa细胞、人乳腺癌MDA-MB-435细胞生长的影响。方法:采用当归水煎液和CO2超临界提取的当归油,分别与体外培养的人前列腺癌PC3细胞、人宫颈癌HeLa细胞、人乳腺癌MDA-MB-435细胞作用48 h后,用磺酰罗丹明B(SRB)染色方法检测细胞活性。结果:当归水提物(500-1 000μg/ml生药)与对照组相比,能够显著升高人宫颈癌HeLa细胞、人乳腺癌MDA-MB-435细胞的OD值(P〈0.05),对人前列腺癌PC3细胞系列未见明显影响。超临界CO2提取的当归油,在3个细胞系列中与对照组相比能够明显降低OD值(P〈0.01),并在0-250μg/ml范围内呈剂量效应关系。对3个肿瘤细胞系半数抑制浓度分别是218.24μg/ml、281.80μg/ml、285.60μg/ml抑制率最高可达到67%。结论:当归水提物有刺激人宫颈癌HeLa细胞、人乳腺癌MDA-MB-435细胞增殖的作用。当归油能够明显地抑制人前列腺癌PC3细胞、人宫颈癌HeLa细胞、人乳腺癌MDA-MB-435细胞增殖。结果提示,当归中能够刺激肿瘤细胞增殖的化合物可能存在于当归水溶性组分中。
Objective:to observe the growth-modulating effects of different extracts of Angelica sinensis Methods: with sulfonylrhodamine B (SRB) assay, in vitro cultured human prostate cancer PC3 cell line, human cervical carcinoma HeLa cell line and human breast cancer MDA-MB-435 cell line were used to evaluate the proliferation effect of the water extract and the essential oil extracted from Angelica sinensis. Resuits: the water extract of Angelica sinensis (500-1000μg/ml) could increase HeLa cells and MDA-MB- 435 cells' figures of OD compared with control (P〈0. 05). Whereas, the essential oil of Angelica sinensis distilled by supercritical-CO2 fluid extraction could obviously reduce the figure of OD (P%0.01), and the effect might be in a dose-dependent manner from 50μg/ml to 250μg/ml. The inhibitory rate could reach 67%. Conclusion: the water extract of Angelica sinensis could stimulate the growth of HeLa cells and MDA-MB-435 cells. The essential oil of Angelica sinensis could inhibit the growth of three kinds of the tumor cells in a dose-dependent manner. These results implicated that the cancer-stimulating component of Angelica sinensis might exist in the water extract.
