不同电刺激对卒中后大鼠神经前体细胞增殖的影响 被引量:2

Effect of different electrostimulation on the proliferation of neural precursor cells in experimental rats with stroke

摘要 目的探讨不同部位电刺激治疗对脑卒中后大鼠神经前体细胞增殖的作用。方法选择2~3月龄,体质量80~120g雄性SD大鼠95只,随机分为梗死灶周围电刺激组(A组)、肢体电刺激组(B组)、对照组(C组)和假手术组(D组)。建立大脑中动脉闭塞模型,在术后24h开始对两电刺激组大鼠头皮和肢体电刺激治疗。在梗死后7d、14d、21d、28d以横木行走试验(BWT)进行神经功能评分,同时进行室管膜和室管膜下5-溴脱氧尿嘧啶核苷(Brdu)的免疫组织化学检测。结果 A组及B组缺血后室管膜Brdu的表达与C组比较在7、14d差异均有统计学意义(P<0.05),MCAO后7d二者相比Brdu在室管膜和室管膜下区的表达差异有统计学意义(P<0.05);MCAO后21dA组Brdu的表达和C组差异有统计学意义(P<0.05),但此时B组和C组差异无统计学意义(P>0.05);在MCAO后第1天及第28天三者之间的Brdu均有表达但差异无统计学意义(P>0.05)。结论电刺激促进MCAO后神经干细胞的增殖,但梗死灶周围头皮电刺激治疗较肢体电刺激治疗引起的神经干细胞增殖明显。 Objective To explore the effect of different electric stimulation on the neural precursor cells in experimental rats with cerebral ischemia. Methods Ninety-five healthy male adult Sprague-Dawley rats (2-3 months old) were randomly divided into infarction focus surrounding ES therapy group (group A), limb ES therapy group (group B), control group (group C) and sham operation group (group D). The middle cerebral artery occlusion model was duplicated with suture methods, then we used beam walking test(BWT) to determine the motor skill of the rats and each cerebral sample was stained with HE and then detected the indexes of immunohistoehemistry: Brdu(5-bromo 2-deoxy-uridine), which was used to evaluate proliferation and differentiation of neuro-precursor cell in the location of infarction. Results Compared with group A and group B, the expression of Brdu in ventricular zone in group C was significantly different at the 7th day and 14th day after MCAO operation (P 〈0.05). Compared with group A, the expression of Brdu in ventrieular zone in group B was significantly different in the 7th day after MCAO operation (P〈0.05), and at 21st day the difference was still exist (P〈0.05), but no difference between group B and group C (P〉0.05). There were no differences among group A, B and C at 1st day and 28th day (P〉0.05). (2onclusion ES can promote the proliferation of neural stem cells (NSCs) after MCAO, however, the effect of ES in the scalp surrounding infarction focus is more effectively than that in the point of limb.
