摘要
通过在培养基中添加底物三丁酸甘油酯,从中、高温大曲中初步筛选到产酯酶细菌15株。以乙酸对硝基苯酯为底物,其中12#菌株表现出最高催化活力其酶活为0.83U/mL;以乙醇、己酸为底物,其中10#菌株表现出最高催化活力其酶活为6.7U/mL。对上述两种测定方法中相对酶活较高的菌株恒温振荡培养(36℃、150r/min)72h后取其发酵液离心(12000r/min,4℃)20min进行气相色谱分析,结果表明这些菌株都能产生一定量的酸类、醇类和酯类物质。
Through adding glyeeryl tri-n-butyrate in the culture me dium, 15 strains producing esterase are obtained after the primary screening in medium and high temperature Daqu. Then using 4-nitrophenyl acetate as substrate, the strain 12# shows the highest catalytic activity of 0.83U/ml; using ethanol and caproic acid as substrate, the strain 10# shows the highest catalytic activity of 33.5U/ml. The two selected strains with relatively higher esterase activity are cultured for 72h at 36℃ with shaking by 150r/min, and the fermented supernatants were centrifuged in 12000r/min at 4℃ for 20min and analysed by GC. The results show that these strains can produce certain amount of acids, alcohol and esters.
