摘要
采用AFLP(扩增性片段长度多态性)分子标记技术对23种37份栽培鸢尾属植物种质资源进行遗传多样性分析,将筛选出的4对AFLP分子标记引物对材料进行扩增,共得到205个位点,其中177个为多态位点,多态位点百分率为86.3%;37份材料之间的遗传相似系数在0.23~0.97之间,平均为0.81.通过遗传相似系数计算,样品总体上亲缘关系非常接近,而白花紫苞鸢尾与其他物种有明显差异.本文还利用UPGMA(非加权算术平均法)进行样品聚类,分析了包括23个种的37份样品之间的遗传关系.
Using AFLP(amplified fragment length polymorphism) molecular markers, we studied the genetic relationship of 37 cultivated Iris materials. Four pairs of primers were used and a total of 205 bands were produced, of which 177 bands were polymorphic. The percentage of polymorphic bands was 86.3~; the genetic similarity among all materials was ranged from 0.23 to 0.97, with an average of 0.81. Overall, the analyses of genetic similarity indi- cated that there was less differentiation among the studied materials except for I. ruthenica f. leucantha was greatly different from the others. In this study, the UPGMA(unweighted pair-group method with arithmetic means) method was also used to analyze the genetic relationship among the collections.
