摘要
目的:探讨苦参碱(MA)对阿霉素(ADM)抗肿瘤作用的影响,以期为乳腺癌的治疗提供新的靶点。方法:MTr法分别检测不同浓度(0,0.125,0.25,0.5,1.0,2g/L)MA、不同浓度(0,0.625,1.25,2.5,5,10mg/L)ADM以及ADM(0.4mg/L)与MA(0.5g/L)合用对乳腺癌细胞株MCF-7的增殖抑制作用。溴化丙啶(PI)单染检测MA(0.5g/L)联合ADM(0.4mg/L)诱导MCF-7细胞凋亡的影响。结果:随着药物作用时间以及药物浓度的增加,MA、ADM以及两种合用对MCF-7细胞增殖的抑制作用逐渐增强。0.5g/LMA诱导MCF-7细胞凋亡率低,仅为12.58%,0.4mg/LADM诱导MCF-7细胞凋亡率为35.12%;而0.5g/LMA与0.4mg/LADM联合使用诱导MCF-7细胞凋亡率为68.11%,明显高于ADM或MA单独使用。结论:MA、ADM及二者联用对MCF-7细胞的增殖抑制作用具有时间依赖性和剂量依赖性。MA本身诱导MCF-7细胞凋亡率低,但它能增加ADM诱导MCF-7细胞的凋亡率。MA可作为抗癌药的化疗增敏剂。
Objective:To determine whether matrine (MA) could synergize with chemotherapeutic agent Adriamycin(ADM) which was commonly used in breast cancer treatment in order to find new targets for human breast cancer chemotherapy. Methods: MCF - 7 breast cancer cells were incubated with different concentrations of ADM (0.625, 1.25, 2.5, 5.0 and 10 m9/L) with or without MA (0,0. 125,0.25,0.5,1.0,2 g/L). Cell viability was measured by the MTT assay. MCF- 7 breast cancer cells were treated with MA(0.5 g/L) and ADM(O. 4 mg/L). Then apoptotic cells were measured by propidium iodide (PI) staining in Flow Cytometry (FCM). Results:The pro- liferation of MCF - 7 breast cancer cells could be inhibited in different concentration of MA combined with ADM or alone . The inhibition was enlarged with the increasing concentration of drugs and prolonged with increasing time of the cells exposed to drugs. Apoptosis of MCF - 7 breast cancer cells was induced by MA, ADM and MA + ADM . The apoptosis rates were 12.5% , 35.12% and 68.11% respectively. Combination of MA and ADM induced a further increase of apoptosis rate. Conclusions : The proliferation of MCF - 7 breast cancer cells can be inhibited by MA, ADM and MA + ADM . This inhibition shows the dose - and exposing time - dependence. The apoptosis rate of MCF - 7 breast cancer cells induced by MA is low, but it can enlarge the apoptosis rate induced by ADM.
