摘要
目的探讨β-榄香烯对多发性骨髓瘤(MM)的白细胞介素-6(IL-6)表达的影响,以期了解β-榄香烯抗MM的分子机制。方法人MM细胞系RPMI 8226与不同浓度的β-榄香烯作用,以酶联免疫吸附试验(ELISA)法检测MM细胞培养上清中IL-6的浓度,逆转录聚合酶链反应(RT-PCR)检测IL-6基因表达。结果不同浓度的β-榄香烯作用RPMI 8226细胞48h后,培养上清中IL-6浓度与对照组相比,差异有统计学意义(P<0.01)。RT-PCR检测结果表明,不同浓度的β-榄香烯作用于RPMI 8226细胞48h,RPMI 8226细胞的IL-6基因mRNA表达强度与对照相比均明显降低。结论β-榄香烯能明显减少MM细胞IL-6的自分泌,抑制RPMI 8226细胞的IL-6基因表达。提示β-榄香烯降低IL-6表达可能是其抗MM的机制之一。
Objective To investigate the effects of β-elemene on the expression of interleukin(IL)-6 of in human multiple myeloma(MM) cells.Methods In order to evaluate the concentrations of IL-6 and the gene expression of IL-6,the supernatant of RPMI 8226 cells were measured by ELISA and RT-PCR methods after treated with different concentrations of β-elemene.Results The concentrations of IL-6 on the supernatant of RPMI 8226 cells were significantly decreased compared with control.The expression of IL-6 gene were significantly reduced after treatment with β-elemene.Conclusion β-elemene can significantly inhibit the secretion and the expression of IL-6 in RPMI 8226 cells,which seems to be one of mechanisms of anti-MM effects of β-elemene.
