摘要 目的:研究从温郁金中提取的β-榄香烯对人骨髓瘤RPMI-8226细胞增殖、周期和凋亡的影响。方法:采用MTT法检测细胞增殖;AnnexinV/PI双标流式术检测细胞周期、细胞凋亡;Hoechst33342/PI双染荧光显微镜观察β-榄香烯处理后骨髓瘤RPMI-8226细胞形态学变化。结果:β-榄香烯对RPMI-8226骨髓瘤细胞生长具有显著抑制作用,呈浓度和时间依赖性。与对照组相比G0/G1细胞比例显著升高,而S、G2/M细胞比例降低。β-榄香烯作用48h可诱导骨髓瘤细胞凋亡,凋亡率随药物浓度而递增。结论:β-榄香烯可有效抑制人骨髓瘤细胞增殖;其抑制作用与细胞周期阻滞和细胞凋亡激活有关。 AIM: To investigate the effects of β-elemene,extracted from curcuma wenyujin,on proliferation, cell cycle and apoptosis of human multiple myeloma RPMI-8226 cells. METHODS: The effect of β-elemene on the growth of human multiple myeloma RPMI-8226 cells was studied through MTT assay,cell cycle and apoptosis was studied by combined Annexin-V protein iodide staining,The morphological changes was studied by combining Hoechst33342-PI staining. RESULTS: β-elemene inhibited the proliferation of RPMI-8226 cells in a time-and dose- dependent manner. Compared with the control cells,the proportions of the RPMI-8226 cells in the G0 /G1 phase rose,and the proportions of the RPMI-8226 cells in the S and G2 /M phases fell decreased. RPMI-8226 cells treated with β-elemene for 48 h induced apoptosis in a dose-dependent manner. CONCLUSION: β-elemene is able to inhibit the proliferation of RPMI-8226 cells by arresting the cell cycle and inducing the cell apoptosis.
