摘要
目的研究纤维素酶酶解生黄芩药材的最佳酶解条件和黄芩素的最佳提取工艺。方法采用高效液相色谱法测定黄芩素含量,以提取率为指标,分别考察浸提倍数、加酶量、pH值、温度、时间对纤维素酶酶解反应的影响,采用正交实验对乙醇浓度、提取温度、提取时间进行考察,并分别考察乙醇用量和提取次数对黄芩素提取率的影响。结果确定了纤维素酶酶解黄芩药材的最佳工艺为:浸提倍数20,加酶量20 U/g药材,pH 4.4,温度50℃,时间8 h。乙醇提取最佳工艺为:加热温度100℃,回流提取时间120 min,乙醇浓度60%,乙醇提取倍数50倍,提取次数为:2次。结论与传统的水煎法及单纯的醇提法相比,纤维素酶酶解反应法使黄芩素的提取率显著提高,通过正交实验优化提取工艺使黄芩素提取率达10%以上。
Objective To s the optimal extraction condition of tudy the optimal cellulolysis condition of raw Scutellaria baicalensis Georgi and baicalein in Scutellaria bacalensis . Methods High performance liquid chromatography method was used to determine the content of baicalein in Scutellaria baicalensis. Five factors were studied on enzymatic extraction of celluase. The orthogonal experiment was adopted to study the effect of the concertration and amount of alcohol, times of refluxing, duration of refluxing and temperature on the extraction rate of baicalein. Results The optimum cellulolysis conditions were determined as follows:20 U of enzyme for 1 g raw material, pH 4.4, the ratio of buffer to raw material 20, temperature 50℃, and reaction time 8 h. The optimum extraction procedure of baicalein was adding 50 times the amount of 60% alcohol, refluxing and extracting twice ,2 h each time, at the temperature of 100℃. Conclusion Compared with the traditional water extraction and alcohol extraction, cellulolysis reaction can increase the extraction yield of baicalein greatly. The extraction rate of baicalein in Scutellaria bacalensis can exceed 10% under the best condition.
