摘要
目的:探讨大黄素对人白血病细胞HL-60增殖及凋亡的影响。方法:应用MTT法检测大黄素对HL-60细胞增殖的影响;应用流式细胞术、DNA琼脂糖凝胶电泳观察和检测细胞凋亡;蛋白印迹法(Western-Blot)检测Bcl-2蛋白表达水平的变化。结果:在5~20mg/L浓度范围内,大黄素对HL-60细胞增殖的抑制作用呈时间剂量依赖性20mg/L作用72h后,抑制率分别达到(19.8±2.7)%,(58.8±4.4)%和(73.8±5.7)%;流式细胞术检测发现:大黄素5、10和20mg/L作用24h后,细胞凋亡率分别为(15.5±3.4)%、(56.0±3.4)%和(71.1±5.0)%,与对照组(1.01±0.06)%有显著性差异(P<0.05);DNA琼脂糖凝胶电泳显示典型的梯形条带。大黄素作用24h后,Bcl-2蛋白的表达水平随药物波度增加呈逐渐下降趋势。结论:大黄素能够抑制HL-60细胞增殖,并诱导其凋亡。Bcl-2基因可能参与了大黄素抑制HL-60细胞增殖和诱导凋亡的过程。
Objective : To investigate the effect of emodin on inducing HL - 60 leukemia cells of proliferation inhibition and apoptosis. Methods : HL -60 cells were exposed to emodin at different dosages. Growth inhibition was detected by MTT assay, and cell apoptosis was detected by flow cytometry and DNA agarose gel electrophoresis. The expression of bcl - 2 was detected by Western - blot. Results : The results showed that when HL - 60 ceils were treated with 5 - 20mg/ L of emodin, the proliferation of HL - 60 cells was obviously inhibited in dose - dependent and time dependent manner. The proliferation rate of HL - 60 cells treated with emodin for 72 hour were ( 19.8 ± 2.7) %, (58.8± 4.4) % and (73.8 ±5.7)%. 24 hours after exposure to 5 ,10,20mg/L emodin, the apoptotic rates of HL -60 were respectively ( 15.5 ± 3.4) %, ( 56.0 ± 3.4) % and ( 71.1 ± 5.0 ) %, and were statistically significant when compared with the control group (1.01 ± 0.06)% ( P 〈 0.05 ). The typical DNA ladder on agarose gel electrophoresis for analysis of cellular apoptosis were significantly appeared in emodin treated HL -60 cells. The bcl -2 protein expression on HL -60 cells were decreased after emodin treated. Conclusion : It is concluded that the emodin can inhibit the proliferation of HL -60 cells and induce the apoptosis in HL -60 cells, Bcl -2 may be involved in this process.
