正交实验法优选大青叶中靛蓝和靛玉红的提取工艺 被引量:7

The optimization of extracting indigo and indirubin from Dyers woad leaves by orthogonal experiment

摘要 探讨了有机溶剂提取大青叶中靛蓝和靛玉红的生产工艺。考察了浸提法、回流法、超声波法和微波法4种提取方法,结果表明,回流法提取效果最好。采用单因素和正交实验考察了甲醇浓度、水浴温度、提取时间、料液比和提取次数对靛蓝和靛玉红得率的影响,最佳提取工艺为:80%甲醇溶液,水浴66℃,料液比1:30,每次提取1.0h,提取4次。在此优化条件下,大青叶中靛蓝和靛玉红的得率分别达到2.088mg/g和0.370mg/g。 The extract technology of indigo and indirubin from Dyers woad leaves was investigated in this paper.Using four extracting methods,such as organic reagent extraction,heating circumfluence extraction,ultrasonic extraction and microwave extraction,indigo and indirubin were extracted by methanol solvent.Among them,heating circumfluence extraction method was the best method.Single factor and orthogonal design L16(45) in the experiment of heating circumfluence extraction were chosen to research the best procedure.The optimum conditions were:80% methanol solution,66 ℃ water bath temperature,1:30 of material and liquid ratio,1.0 h extraction time and extracted 4 times.The yield of indigo and indirubin under the optimum conditions were 2.088 and 0.370 mg/g,respectively.
