摘要 目的研究IL - 2基因修饰人肿瘤细胞生物学行为是否因辐射而发生变化。 方法分别从细胞形态结构、MHC分子表达、整合的基因IL - 2基因是否丢失、能否保持mRNA表达、能否分泌表达生物活性的IL - 2等方面进行了实验。结果未见整合的IL - 2基因丢失 ,能表达IL - 2mRNA且在一定时间内持续分泌具有生物活性的IL - 2 ;细胞膜表面MHC分子无明显改变 ;细胞形态结构无明显变化。结论经60 Co辐射修饰的肿瘤细胞致瘤性消失 ,但在一定时间内能分泌IL - 2 ,这为开展基因工程“瘤苗”的临床应用提供了依据。 Objective To study the biological behavior changes in gene-modified tumor cells' after irradiation with 60 Co. Methods We examined the cell morphaogical structure, expression of MHC molecule, integration and expression of the IL-2 gene of the IL-2 gene transduced tumor cells and their tumorigenicity after irradiation with 60 Co. Results The data showed:1.the integrated IL-2 gene was not lost and rearranged, a IL-2 mRNA still normally expressed and the biological active IL-2 was persistently secreted for a certain period; 2. the MHC molecules expressed on the cell membrane did not change significantly; 3. the tumorigenicity of the modified tumor cell was eliminated; 4. the cell morphogical structure had not changed significantly. Conclusion The tumorigenicity of the transduced tumor cell after irradiation 60 Co was eliminated, but persisted to secret IL-2 for a certain period. All these data provide logical basis for the clinical uses of cytokine gene engineering tumor cells as tumor vaccine.
