摘要
目的:观察脑缺血后iNOS基因表达情况。方法:在建立SHRMCAO局灶性脑缺血模型基础上,采用半定量RT-PCR技术观察脑缺血后不同时相iNOSmRNA表达水平变化。结果:显示脑缺血后1h、4h缺血区iNOSmRNA基因无明显改变,12h时iNOSmRNA基因表达升高(P>0.05),24h、48h后iNOSmRNA基因表达明显增加(P<0.01)。结论:脑缺血后中、晚期iNOS基因表达增加,其产生的NO可能参与脑缺血后期的继发性病理改变。
Aim: The changes of the gene expression of iNOS mRNA after cerebral ischemia were observed. Methods: On the basis of the model of the middle cerebral artery occlusion in SHR, the gene expression of iNOS mRNA were determined by semi-quantity RT-PCR method. Results: It was showed that the gene expression of iNOS mRNA in infarct area in 1h and 4h groups was higher than control group, but there was no significant difference (P>0.05), and it in 24h and 48h groups after MCAO was significantly increased compare to control group (P<0. 01). Conclusion: It indicates that the gene expression of iNOS mRNA increased at the moderate and late stage after MCAO, and nitric oxide produced by iNOS might take part in the secondary pathological changes at late stage of cerebral ischemia.
