摘要
目的通过体外实验探讨压力刺激对大鼠"足三里"穴筋膜组织细胞合成释放肿瘤坏死因子-α(TNF-α)和干扰素-β(INF-β)的影响,为腧穴"扶助正气"的治疗作用提供细胞生物力学方面的实验依据。方法体外培养大鼠"足三里"穴筋膜组织细胞,对细胞实施免疫组织化学鉴定后,实施100 kPa和200 kPa压力刺激,检测细胞外培养液INF-β和TNF-α含量的变化。结果 "足三里"穴筋膜组织细胞Keratin染色阴性,Vimnetin染色阳性,200 kPa压力刺激后,培养液中TNF-α含量增加,INF-β含量降低,差异均有统计学意义。结论 "足三里"穴筋膜组织细胞主要是成纤维细胞,压力刺激对筋膜组织成纤维细胞TNF-α和INF-β合成释放的调节,可能是腧穴接受针灸推拿治疗后特异性发挥"扶助正气"作用的细胞生物力学原理之一。
Objective To supply experimental bases from the view of cellular biomechanics for the treatment of acupuncture points "strengthening body resistance" by exploring the effect of stress on synthesis and release of INF-β and TNF-α by rat "Zusanli" fibroblast cells in vitro.Methods Rat "Zusanli" Point fascia cells were cultured in vitro and identified through immoral histochemistry(SABC).100 kpa and 200 kPa strain was taken on cells,respectively.INF-β and TNF-α in extracelluar nutrient solution were examined. Results Keratin staining of "Zusanli" Point fascia cell was negative and Vimnetin staiming was positive.Under 200 kPa pressure,the content of INF-β decreased,that of TNF-α was increased,and there had statistical difference. Conclusion Fibroblast cells are the main ingredient of "Zusanli" Point fascias.Pressure stimulation on regulation of acupoints fascias may be one of the celluar biomechanical principles of the special effect of "strengthening body resistance" after being treated by acupuncture and massage.
