摘要
以DPPH清除率为指标,研究甜菊叶提取物清除DPPH的能力。在单因素试验基础上,运用响应面法确定甜菊叶提取物的最佳提取条件为提取温度60.4℃、提取时间2.98h、水料比15.4:1。在此条件下得到的甜菊叶提取物对DPPH的清除率最大,88.78%;其清除DPPH的半数有效剂量EC50为32.3μg/mL,自由基清除能力AE(1/EC50)为0.0310。研究表明,甜菊叶提取物具有一定的清除DPPH的能力。
The DPPH scavenging capacity of Stevia rebaudiana Berton extract was studied in this paper by using the clearance of DPPH as the index.Based on the results of single factor experiments,the extraction conditions of Stevia rebaudiana Berton were optimized by using the response surface methodology.The results showed that the optimum extraction conditions were temperature 60.4 ℃,extraction time 2.98 h,and water/material ratio 15.4:1.Under these conditions,the DPPH scavenging rate,the median effective dose(EC50) and the radical scavenging activity AE(1/EC50) of the Stevia rebaudiana Berton extract reached 88.78%,32.3 μg/mL and 0.0310 respectively,indicating the DPPH scavenging capacity of Stevia rebaudiana Berton extract.This study provided a importantly theoretical foundation for development and application of Stevia rebaudiana Berton natural antioxidant.
