摘要
目的探讨细胞周期蛋白(cyclinB1)、细胞周期蛋白依赖性激酶(CDK1)及细胞周期蛋白依赖性激酶抑制物(CDKIs-P27)在宫颈鳞癌组织中的表达及其意义。方法采用免疫组织化学SP法检测18例正常宫颈、19例宫颈上皮内瘤变(CIN)、56例宫颈鳞状细胞癌组织中cyclinB1、CDK1及P27蛋白的表达。结果 cyclinB1、CDK1在宫颈鳞癌及CIN中的阳性表达率明显高于在正常宫颈组织中(P<0.05),而P27在正常宫颈组织中呈高表达,在宫颈鳞癌及CIN组织中显著降低(P<0.05)。cyclinB1、CDK1在宫颈鳞癌组织中的表达与肿瘤组织学分级有关(P<0.05),P27表达与组织学分级、淋巴结转移有关(P<0.05)。宫颈鳞癌组织中cyclinB1与P27的表达、CDK1与P27的表达分别呈负相关关系(P<0.05)。结论 cyclinB1、CDK1及P27的异常表达在宫颈鳞癌的发生发展、转移中起重要作用。联合检测这些指标有助于判断宫颈鳞癌的生物学行为及转移潜能。
Objective To discuss the protein expression and significance of cyclinB1,CDK1 and P27 in cervical squamous cell carcinoma.Methods Expression levels of cyclin B1,CDK1 and p27 in surgical specimens from 18 cases of normal cervical tissue,19 cases of cervical intraepithelial neoplasia(Ⅱ,Ⅲ level) tissue,and 56 cases of cervical squamous cell carcinoma.Results The positive rate of cyclinB1,CDK1 in cervical squamous cell carcinoma and CIN were higher than that in normal cervix tissue,while the positive rate of P27 was very high in normal cervix tissue,but was significantly lower in the tissue of cervical squamous cell carcinoma and CIN.The differences were statistically significant(P〈0.05).The positive rate of cyclinB1,CDK1 in cervical squamous cell carcinoma was significantly associated with histological grade(P〈0.05).The positive rate of P27 in cervical squamous cell carcinoma was associated with lymph node metastasis.There was inversely correlation between positive expression of CyclinB1 and p27(r=-0.331,P〈0.05).There was inversely correlation between positive expression of CDK1 and p27(r=-0.344,P〈0.05).Conclusions The abnormal expression of cyclinB1,CDK1 and P27 in cervical squamous cell carcinoma may play some important roles in conjunction with a function in the growth and progression of cervical squamous cell carcinoma.There will have contribution to evaluate the biology characteristic and metastatic potential in cervical squamous cell carcinoma with consociation examination of them.
