摘要
目的建立自身免疫性慢性非细菌性前列腺炎大鼠模型,研究其前列腺平滑肌细胞内游离Ca2+浓度的变化及意义。方法将SD大鼠分为正常对照组和炎症组,采用正常SD大鼠前列腺蛋白提纯液及弗氏完全佐剂(FCA)诱导大鼠自身免疫性炎症,两组分别进行体外培养前列腺平滑肌细胞(PSMCs)并纯化,用Fluo 3-AM Ca2+探针结合流式细胞术对PSMCs内游离Ca2+浓度进行测定,比较两组间差异。结果流式细胞术检测波长为525 nm时测定两组大鼠前列腺平滑肌细胞内Ca2+浓度荧光强度值,结果显示炎症组荧光强度值明显高于正常对照组。结论慢性前列腺炎可能导致前列腺平滑肌细胞内游离Ca2+浓度升高,使钙稳态产生波动继而影响到前列腺及细胞的各项生理功能,PSMCs内游离Ca2+浓度在发病过程中具有一定作用。
Objective To establish an autoimmune model of chronic abacterial prostatitis(CAP) of SD rat and to investigate the changes and significant of intracellular calcium ion concentration of smooth muscle.Methods SD rats were divided into two groups,and the model group of CAP was established by purified prostate protein and Freund′s complete adjuvant(FCA) injection.Both of the prostate smooth muscle cells(PSMCs) were cultured in vitro and purified.The intracellular calcium ion concentration of PSMCs was assayed by the Fluo 3-AM calcium detection and flow cytometry.Results Compared with the control group,the fluorescence intensity value of model group was higher with an emission wavelength of 525 nm.The fluorescence intensity value was proportional with the intracellular calcium ion concentration.Conclusion The intracellular calcium ion concentration of PSMCs is higher in prostatitis group which may disturb the balance and affect the physiological function of the PSMCs,and play a role of pathogenesis of the disease.
