摘要
目的:观察直刺、沿皮刺三阴交穴对寒凝证类痛经大鼠子宫组织及脊髓腰膨大中缩宫素(OT)及其受体(OTR)的影响,探讨不同刺法即刻镇痛效应的部分机制。方法:64只SD雌鼠随机分为盐水组、模型组、直刺组、沿皮刺组,每组16只。采用全身冷冻加雌二醇注射法制备寒凝证类痛经模型。直刺组与沿皮刺组分别直刺或沿皮刺三阴交穴,留针20min。记录各组大鼠的扭体反应及子宫收缩程度。采用ELISA法、Real Time-PCR法检测大鼠子宫组织及脊髓腰膨大中OT含量、OTR mRNA表达。结果:与盐水组比较,模型组扭体评分、子宫收缩波个数、活动度均显著升高(P<0.01)。与模型组比较,直刺组、沿皮刺组均显著降低扭体评分、子宫收缩波个数及活动度(P<0.01,P<0.05)。与盐水组比较,模型组OT含量、OTR mRNA表达在子宫组织显著升高(P<0.01),在脊髓显著降低(P<0.01)。与模型组比较,直刺、沿皮刺组均显著降低子宫组织OT含量(P<0.01)、OTR mRNA表达(P<0.05),升高脊髓OT含量(P<0.01)。直刺组与沿皮刺组间各指标均无显著差异。结论:直刺、沿皮刺三阴交穴对类痛经大鼠均显示出即刻镇痛效应,其机制可能与针刺对子宫与脊髓OT、OTR的不同调节有关。
Objective:To observe the effects of perpendicular needling and transverse needling at Sanyinjiao(SP6)on oxytocin(OT)and its receptor(OTR)in uterus tissue and spinal cord waist enlargement of dysmenorrhea rats with cold stagnation syndrome,and to explore underlying mechanisms.Methods:A total of 64 SD female rats were randomly divided into saline group,model group,perpendicular needling group and transverse needling group,16 rats in each group.The rat model of dysmenorrhea with cold stagnation syndrome was prepared by systemic freezing and estradiol injection.In the acupuncture intervention groups,SP6 was needled perpendicularly or transversely,and the needles were retained for 20min.The writhing reaction and uterine contraction degree of rats in each group were recorded.ELISA and Real Time-PCR were used to detect OT content and OTR mRNA expression in uterine tissue and spinal cord waist enlargement.Results:Compared with saline group,the torsion score,the number of uterine contractions and the uterine motility of model group were significantly increased(P<0.01).Compared with the model group,the torsion score,the number of uterine contractions and the uterine motility were significantly reduced in the perpendicular and transverse needling groups(P<0.01,P<0.05).Compared with saline group,OT content and OTR mRNA expression in model group were significantly increased in uterine tissue(P<0.01)and significantly decreased in spinal cord(P<0.01).Compared with the model group,the OT content(P<0.01)and OTR mRNA expression(P<0.05)in uterine tissue were significantly decreased in both the perpendicular and transverse needling groups;while the OT content in spinal cord were increased(P<0.01).There were no significant differences in all measures between the perpendicular and transverse needling groups.Conclusion:Perpendicular needling and transverse needling at Sanyinjiao(SP6)show immediate analgesic effect on dysmenorrhea rats with cold stagnation syndrome,and its mechanism may be related to different regulation of OT and OTR o
