摘要
目的研究大骨节病的可疑致病因子T-2毒素对原代培养的兔软骨细胞增殖的影响,探讨大骨节病的发病机制。方法分别用含0,1,10,20,100μg/L T-2毒素的DMEM/F-12培养基体外培养兔软骨细胞5 d,采用四氮甲基唑蓝法每天检测软骨细胞的存活率。结果软骨细胞的生长抑制程度与T-2毒素呈浓度依赖和时间依赖关系;T-2毒素的抑制作用在第5天时接近饱和。结论 T-2毒素确能抑制软骨细胞的生长,可能是大骨节病的重要病因。
Objective To observe the effect of trichothecene mycotoxin(T-2 toxi n),a possible pathogenic factor of Kashin-Beck disease(KBD),on proliferation of Zelanian rabbits'chondrocyte in vitro.Methods Rabbit chondrocytes cultured in vitro were treated with T-2 toxin at different concentrations(0,1,10,20,100 μg/L) for 5 d.Cell viability was measured by methyl thiazolyl tetrazolium assay everyday.Results Dose/time-dependent effect of T-2 toxin was found in chondrocyte viability within a certain range of concentration(1-100 μg/L) fro m the first to fifth day.The inhibitory effect of T-2 toxin on chondrocytes was up to maximum on the fifth day.Conclusion T-2 toxin inhibitted the growth of r abbit chondrocytes and maybe the key role in pathogenesis of Kashin-Beck diseas e.
