摘要
目的探讨4gLib伤性脑梗塞(traumaticcerebralinfarction,TCI)与血浆纤维蛋白原(Fg)Bβ-455G/A基因多态性及其水平的相关性。方法2()03年i月至2011年3月对我院71例TCI患儿及40例外伤无TCI患儿末梢血标本应用PCR限制性片段长度多态性(PCR-RFLP)技术,检测血浆纤维蛋白原-455G/A基因,应用CA-530全自动凝血分析仪检测血浆纤维蛋白原水平。结果TCI患儿Bβ-455G/A位点的GG基因型和G等位基因出现频率73.2%和83.8%与对照组45.0%和63.8%比较明显增高;OG型发生TCI的危险性较AA型高约4倍(OR=3.967,95%CI:I.17~10.896);Fg水平TCI组(65.3±17.7)mg/L较对照组(32.7±10.9)mg/L明显增高。结论&JLTCI的发生可能与纤维蛋白原Bβ-455G/A基因多态性及其水平相关;β纤维蛋白原-455A/G位点GG基因型可能是小儿TCI的危险因子;动态监测Fg水平,对于早期诊断TCI有一定意义。
Objective To investigate the relationship between children with traumatic cerebral infarction (TCI) and plasma fibrinogen gene polymorphism and level. Methods Use the method of PCR-RELP to detect the -455G/A gene type of plasma fibrinogen in 71 cases children with traumatic cerebral infarction and in 40 cases children without traumatic cerebral infarction. The levels of plasma fibrinogen were detected by CA-530 automatic blood analyzer in hospital from January 2003 to March 2011. Results Genetype of GG and allele of G in B13-455G / gene site in group with traumatic cerebral infarction (73.24% and 83. 8%) had a higher frequency than that in control group (45.0% and 63. 8%). The traumatic cerebral infarction relative risk of GG genotype in Fg- Bβ455G/A site was 4 times that of the AA genotype. The levels of FG in traumatic cerebral infarction group (65.3± 17. 7) mg/L was significantly higher than that in control group (32. 7 ± 10. 9) mg/L. Conclusions The inci- dence of traumatic cerebral infarction in children may be related to the gene polymorphism and levels of fibrinogen gene Bβ-455G/A. GG genotype of Bβ-455G/A may be the a risk factor of traumatic cerebral infarction in children. Dynamic levels of FG is valuable in early diagnosis of traumatic cerebral infarction.
