摘要
目的从分子水平上阐明河南省兰考地区捕获的野生小鼠(LK)与4个小鼠品系(B6,BALB/c,DDK,PWK)之间的遗传差异及遗传关系,从而进一步确定该种野生小鼠的种属及培育野生来源近交系小鼠品系。方法利用25对引物,对野生小鼠及4个小鼠品系进行扩增片段长度多态性(AFLP)分析。结果检测到2035条扩增条带,多态性条带有507条,占总条带的24.91%,并且LK小鼠与PWK、DDK、BALB/c、C57BL/6J的遗传距离指数分别为0.01696、0.02790、0.02729、0.02759。结论 LK小鼠与PWK小鼠品系遗传关系最近,遗传距离指数最小;同时也证明了AFLP技术能够在物种鉴定、遗传背景分析及预测亲本杂交效果等方面具有明显的优越性。
Objective To determine the genetic variation and genetic relationship between the wild mouse captured from Lankao county,Henan province,and four common strains of laboratory mice(B6,BALB/c,DDK,PWK),and to construct a wild-origin inbred line mouse.Methods Totally 25 pairs primers were used to identify DNA polymorphisms that distinguish the five strains of mice by amplified fragment length polymorphism(AFLP).Results AFLP primer combinations detected a total of 2035 amplification bands.There were 507 polymorphic bands,24.91% of total bands.The genetic distance index between LK mice and PWK,DDK,BALB/c,C57BL/6J mice was 0.01696,0.02790,0.02729,and 0.02759,respectively.Conclusions Th results of this study show that LK mouse and PWK mouse have the closest genetic relationship and the genetic distance index between them is the least.It also demonstrates the apparent advantages of AFLP technique in species identification,genetic background analysis and prediction of effects such as hybrid parents.
