D1S80座位的等位基因扩增片段长度多态性分析

摘要 目的:对天津地区112 名无关个体的D1S80 基因座位进行了扩增片段长度多态性分析,检测其等位基因和基因频率。方法: 应用聚合酶链反应,小型聚丙烯酰胺凝胶电泳和银染方法观察长度多态性。结果:发现了26 个等位基因,大小在355~771bp 之间,基因频率为0.0045~0.1384,杂合度为73% 。70 种基因型,个人鉴别力为0.985。对6 个家系23 名相关个体的分析结果符合孟德尔定律。结论: D1S80 基因座位个人鉴别力高,PCR方法简便、灵敏。 Objective: The amplified fragment length polymorphism (Amp-FLP) of locus D1S80 among 112 unrelated TianJin chinese residents was analysed. Methods: This analysis was performed with polyacrylamide mini-gel electrophoresis followed by the silver staining. Results: 26 alleles, ranging from 355 to 771 bp in size and from 0.0045 to 0.1384 in frequency, and 70 genotypes were observed. The heterozygosity was 73% , and the discrimination power (DP) was 0.98 . The study on six families demonstrated that this locus was inherited according to Mendelian law. Conclusion: The technique used here is quite simple, rapid, extremely sensitive, and does not require the use of radioisotopes. It could provide a powerful new tool for the forensic idendification of individuals and paternity testing.
