摘要
目的探讨1,2-二甲基-3-羟基-4-吡啶酮(1,2-dimethyl-3-hydroxy-4-pyridone,deferiprone,DFP)对铝染毒小鼠血清蛋白和肝脏酶活力的影响。方法将50只健康清洁级昆明小鼠按体重随机分为5组,分别为阴性对照(生理盐水)组、铝(200 mg/kg)单独染毒组和低(17.58 mg/kg)、中(35.15 mg/kg)、高(70.30 mg/kg)剂量DFP+铝(200 mg/kg)染毒组,每组10只,雌雄各半。第1~4周,灌胃染毒AlCl3溶液,染毒容量为10 ml/kg,每天1次,每周连续5 d;第5周,灌胃染毒DFP溶液,染毒容量20 ml/kg,每天1次,连续5 d。测定血清中总蛋白(TP)、白蛋白(Alb)含量和γ-谷氨酰转移酶(γ-GT)活力以及肝脏中铝含量和ATP酶、黄嘌呤氧化酶(xanthine oxidase,XO)的活力。结果与阴性对照组比较,铝单独染毒组肝脏铝含量较高(P<0.01),血清TP含量较低(P<0.01),给予DFP后各组均恢复至阴性对照组水平(P>0.05);铝单独染毒组血清Alb含量比阴性对照组降低(P<0.05),给予中、高剂量DFP后恢复至阴性对照组水平(P>0.05);与阴性对照组比较,铝单独染毒组和低、中剂量DFP+铝染毒组肝脏Ca2+-Mg2+-ATP酶活力均较低(P<0.01),XO活力较高(P<0.01),仅高剂量DFP+铝染毒组肝脏Ca2+-Mg2+-ATP酶活力与阴性对照组无差异(P>0.05),低、中、高剂量DFP+铝染毒组XO活力低于铝单独染毒组(P<0.05,P<0.01),且高剂量DFP+铝染毒组与阴性对照组无差异(P>0.05)。结论服用DFP对铝染毒小鼠血清蛋白和肝脏酶活力的改变具有一定的恢复作用。
Objective To investigate the effect of given deferiprone on serum protein and liver enzymes activity in aluminum exposed mice.Methods Fifty healthy clean grade KM mice were randomly divided into 5 groups:negative control(saline)group,single-aluminum treated group(200 mg/kg),low(17.58 mg/kg),moderate(35.15 mg/kg),high(70.30 mg/kg)-dose deferiprone + aluminum group.Every group had 5 female mice and 5 male mice.The mice were treated with AlCl3 solution at the volume of 10 ml/kg through gavage for the first 4 weeks,once a day,for 5 consecutive days.At the fifth week,mice were treated with different doses of deferiprone solution at the volume of 20 ml/kg,once everyday.Then the concentration of TP,albumin(Alb),γ-GT in serum,concentration of aluminum and activity of Na+-K+-ATPase,Ca2+-Mg2+-ATPase,xanthine oxidase in liver tissues were determined.Results Compared with the negative control group,the liver aluminum contents were higher(P0.01) and the concentration of serum TP were lower(P0.01) in single-aluminum treated group,and after mice were treated with deferiprone,they recovered to the level of negtive group(P0.05).The serum albumin concentration of single-aluminum treated group was lower than that of negative control group(P0.05),and the index was recovered after treated with moderate,high-dose deferiprone(P0.05).The activity of liver Ca2+-Mg2+-ATPase in single aluminum,low and medium-dose deferiprone+aluminium groups were lower than that of negative control group(P0.01),and there was no significant difference between high-dose deferiprone+ aluminium group and negative control group(P0.05).The activity of liver XO in single aluminum group and low,moderate-dose deferiprone + aluminum groups was higher than that of negative control group(P0.01),as three doses groups were lower than single aluminum treated group(P0.05,P0.01).No significant differences existed between high dose deferiprone+ aluminium group and negative control group in
