摘要
采用高通量筛选(high throughout screening,HTS)技术,以高胆固醇症细胞模型LXR(肝脏X受体)和代谢综合症细胞模型FXR(胆汁酸受体)、肝癌细胞株HepG2、白血病细胞株HL-60为对象,研究了芹菜素、芹菜苷、3′-甲氧基芹菜苷、芹菜醇提物、芹菜粗黄酮等芹菜黄酮类物质对治疗代谢综合症以及抑制肿瘤细胞增殖的活性。结果表明:活性筛选药物的添加浓度为10μg/mL时,在肝脏X受体模型上芹菜素表现出一定的活性,同时也表现出有利于提高胆汁酸受体模型敏感性的作用;芹菜素和芹菜醇提物样品对HepG2和HL-60均有一定程度的抑制作用,芹菜素和芹菜醇提物抑制HepG2的GI50值分别为3.31μg/mL和66.07μg/mL,抑制HL-60的GI50值分别为1.62μg/mL和44.67μg/mL。
The pharmacologic activity of apigenin, 3 "-methoxy-apigenin, celery ethanol extraction and celery rough flavonoids for curing metabolic syndrome and inhibiting cancer cell multiplication was studied by high throughout screening (HTS) technology with LXR (liver X receptor) , FXR (farnesoid X receptor) model, liver cancer cell HepG2 and leukemia cell HL-60. When the concentration of samples was 10μg/mL, apigenin performed a certain pharmacology activity for LXR receptor and increased sensibility for FXR receptor. Apigenin and celery extraction have certain inhibition effects on HepG2 and HL-60.When the concentration was 100μg/mL, the GIS0 of apigenin and celery ethanol-extraction for inhibiting HepG2 was 3.31μg/mL and 66.07 μg/mL, respectively, and 1.62 μg/mL and 44.67μg/mL for inhibiting HL-60, respectively. Meanwhile apiin has no inhibition effects on HepG2 and HL-60.
