摘要
目的研究芹菜素对人乳腺癌MDA-MB-231细胞血管内皮生长因子(vascular endothelial growth factor,VEGF)表达的影响。方法采用MTT法检测芹菜素(apigenin)对乳腺癌细胞增殖的影响;应用酶联免疫吸附法检测VEGF在MDA-MB-231细胞中的分泌水平;应用RT-PCR法检测MDA-MB-231细胞中VEGFmRNA的表达水平;应用蛋白质印迹方法检测芹菜素对MDA-MB-231细胞中HIF-1α,p-AKT,p-ERK1/2,P53表达的时间依赖性和剂量依赖性效应。结果芹菜素对MDA-MB-231细胞没有明显增殖抑制效应。芹菜素能够抑制MDA-MB-231细胞分泌VEGF,并且降低VEGF mRNA的表达水平。同时,芹菜素抑制了MDA-MB-231细胞中HIF-1α,p-AKT的表达,诱导P53的表达,而对p-ERK1/2的表达没有影响。结论芹菜素能够抑制人乳腺癌细胞VEGF的表达,这种效应可能通过降低HIF-1α的表达来实现。
[Objective] To study the effects of Apigenin on vascular endothelial growth factor (VEGF) in human breast cancer cells (MDA-MB-231). [Methods] MTr assay was used to detect the cell viability inhibitory effect of apigenin on MDA-MB-231 cell. ELISA was used to determine the protein level of VEGF secreted by MDA-MB- 231 cell. RT-PCR was used to detect mRNA levels of VEGF in MDA-MB-231 cell. The protein levels of HIF-1α, p-AKT, p-ERK1/2, and P53 were detected by Western Blotting. [Results] Apigenin did not inhibit the cell viability of MDA-MB-231. Apigenin reduced the secretion of VEGF, and mRNA level of VEGF in MDA-MB-231 cell. Additionally, Apigenin decreased the expression of HIF-1α, p-AKT, and p-ERK1/2, but induced the expression of P53. [Conclusion] Apigenin can inhibit VEGF expression in human breast cancer cells, and this may be achieved through decreaseing HIF-1α.
