铝螯合剂对铝染毒大鼠肝脏中铝的排出和血清蛋白质及胆红素的影响 被引量:2

摘要 目的观察铝螯合剂1,2-二甲基-3-羟基-4-吡啶酮(deferiprone,DFP)对染铝大鼠肝脏中铝的排出及对血清中蛋白质和胆红素含量的影响。方法30只雄性Wistar大鼠按体重随机分为6组,阴性对照组、铝染毒组、DFP预防组及铝+DFP低、中、高剂量组,阴性对照组(给予生理盐水1ml/d,连续4周);铝染毒组及铝+DFP低、中、高剂量组,给予AlCl3·6H2O281.40mg/(kg·d)3周后,第4周分别给予生理盐水1ml/d、DFP13.82、27.44、54.88mg/(kg·d)。预防组给予AlCl3·6H2O281.40mg/(kg·d),4h后给予DFP27.44mg/(kg·d),给药4周。测定血清总蛋白、白蛋白和总胆红素、结合胆红素含量及肝脏中铝的含量。结果铝染毒组肝组织铝含量明显高于阴性对照组(P<0.01),铝+DFP低、中、高剂量组均明显低于铝染毒组(P<0.01),且呈剂量-效应关系。铝染毒组大鼠血清中总蛋白含量显著低于阴性对照组(P<0.01),铝+DFP中、高剂量组则显著高于铝染毒组(P<0.01),且与阴性对照组相比无统计学意义;各组大鼠血清中白蛋白含量比较均无统计学意义;与阴性对照组相比,各组大鼠血清中总胆红素含量均显著增加(P<0.05,P<0.01),而结合胆红素含量比较均无显著性差异。结论DFP通过促进肝脏中铝的排出,恢复肝脏合成蛋白的功能,但对胆红素未产生明显影响。 Objective To investigate the effect of 1,2-dimethyl-3-hydroxypyrid-4-pyridine (deferiprone, DFP) on protein and bilirubin in serum and aluminum in liver of rats exposed to aluminum. Methods Wistar rats were administered with aluminum chloride for 3 weeks. Then they were treated with different doses of DFP[13.82?27.44?54.88 mg/(kg·d)]. The contents of total protein(TP), albumin, total bilirubin(TB) and conjugated bilirubin in serum were detected, and the concentration of Al in liver tissue was measured. Results The aluminum content in liver of the three different dose groups(Al+DFP) was significantly lower than that of the Al group (P<0.01). The TP content in serum of the Al group was significantly lower than that in the negative control(P<0.01). The TP content in serum of moderate and high DFP group were higher than the Al group(P<0.01), while the TP content in serum showed no significant differences between the moderate and high DFP dose groups and negative control. The albumin contents in serum showed no statistical significance among all the groups. Compared with the negative control, the TB contents in serum of the other five groups were significantly higher (P<0.05, P<0.01). The contents of the conjugated bilirubin in serum in all groups showed no significant difference. Conclusion DFP can recover the function of protein synthesis by increasing aluminum output in liver. But the effect of DFP on bilirubin is not observed.
