摘要 以野葛叶为原料,采用水提醇沉法,通过单因素试验和正交试验,以凯氏定氮法测得野葛叶蛋白质含量和提取率,对提取温度、提取时间、溶液pH值、液料比4个因素进行优化研究,确定野葛叶蛋白质提取的最佳方法。结果表明,从野葛叶中提取蛋白质的最佳工艺条件为:提取温度90℃,提取时间120 min,提取溶液的pH值6,液料比1.4∶1(V/W,L∶kg)。 Ethanol subsiding method was adopted to extract protein from Pueraria lobata leaves.Single factor and orthogonal test combined with Kjeldahl method was used to optimize the extracting conditions including extraction temperature,extraction time,solution pH,and liquid to solid ratio.The results showed that the optimum conditions of protein extraction from leaves of Pueraria lobata were extraction temperature,90 ℃;extraction time,120 min;solution pH,6;and liquid to solid ratio, 1.4∶1(V/W,L:kg).
