摘要 [目的]确定藿香叶蛋白质的最佳提取工艺条件。[方法]以风干的藿香叶为原料,分析了料液比、pH值、提取温度、提取时间对叶蛋白质提取率的影响。并在单因素提取试验的基础上,设计了正交优化试验。[结果]提取时间对藿香叶蛋白质提取率的影响不是特别大,其他3个因子的影响顺序为:料液比>pH值>温度。在料液比1∶30,pH值9.5,温度45℃的条件下,提取75min,水溶性蛋白质的提取率最大,达88.54%。[结论]确定了藿香叶蛋白的最佳提取工艺条件,为综合利用东北藿香叶提供了依据。 [Objective] The best extraction process of protein from wrinkled gianthyssop(Agastache rugosa)leaf was determined through the experiment.[Method] The ratio of raw material(naturally dried wrinkled gianthyssop leaf)to liquid,pH value,extraction temperature and time on the yield of extracted protein was analyzed by means of the design of the optimal orthogonal test based on single factor experiment.[Results] The effect of extraction time on the extraction rate of the protein from wrinkled gianthyssop leaf was not particularly large,the order of other three factors affecting the extraction rate was:ratio of raw material to liquid pH value temperature.Under the following condition:the ratio of raw material to liquid,1∶30;pH value,9.5;the temperature 45 ℃;and the extraction time,75 min,the maximum extraction rate of water-soluble protein was as high as 88.54%.[Conclusion] The best extraction condition of the protein from wrinkled gianthyssop leaf was formed,which would provide the basis of the comprehensive utilization of wrinkled gianthyssop leaf.
