摘要
以2009年度参加黄河流域国家棉花区试的57份参试品种为材料,运用筛选确定的22对多态性好、条带清晰的SSR引物对参试品种进行分子标记分析,共检测出145个等位位点,其中106个为多态性位点,平均每对引物4.8个,多态性比率平均达到72.7%,PIC平均值为0.661。利用106个多态性片段构建了这些品种的SSR指纹图谱。遗传聚类结果显示,在相似系数为0.98时,57个区试品种完全区分开;同一地区和同一单位育成的品种在不同程度上聚在一类,说明它们的遗传关系较近。
In this study,57 cotton cultivars in 2009 national regional trails were used as plant materials for DNA fingerprinting.Among 1800 pairs of primers,22 pairs with steady polymorphism were selected.Totally,145 bands were observed,and 106(72.7%) were polymorphic,with an average of 4.8 bands per primer.The mean PIC value of these markers was 0.661,and a DNA fingerprint database for the varieties tested was developed.UPGMA cluster analysis of Genetic similarity showed that the observed 106 polymorphic markers are sufficient to separate all of the 57 varieties from one another at the maximum genetic similar coefficient of 0.98.Cultivars derived from the same district or bred by same institutes were clustered together in different extent,indicating their close genetic relationship.
