摘要
目的探讨E钙黏附蛋白和凋亡相关蛋白Survivin、Fas与胃癌分化程度的关系,及胃癌组织中E钙黏附蛋白、Survivin、Fas间的相关性。方法采用免疫组织化学法检测63例(高、中、低分化)胃癌及22例正常胃黏膜组织中三者的表达。结果 E钙黏附蛋白胃癌中表达率为73%,高于正常胃组织中的64%,Fas在胃癌组织中的表达率为6%,低于正常胃组织中的86%。Survivin在63例胃癌组织中阳性表达87%,明显高于正常组织中的18%,组间相比差异有统计学意义(P<0.05)。E钙黏附蛋白在高、中、低分化癌的阳性表达率依次为45%、33%、5%,Fas在不同分化癌的阳性表达率依次为36%、19%、10%,E钙黏附蛋白、Fas蛋白的表达率在高、中分化癌与低分化癌相比差异有统计学意义(P<0.05)。Survivin阳性表达率由高分化癌的68%到低分化癌的85%有逐渐增强的趋势,但不同分化程度相比差异无统计学意义。E钙黏附蛋白与Fas蛋白在不同分化癌中的表达趋势较一致,但之间无相关性。E钙黏附蛋白、Survivin蛋白在胃癌组织中表达呈正相关(r=0.383,P<0.05);Survivin、Fas蛋白在胃癌组织中表达呈负相关(r=-0.463,P<0.05),未发现三者间有明显相关性。结论 E钙黏附蛋白、Fas在胃癌中表达率低于正常胃组织,且两者与分化程度有关,而Survivin正好相反,从而进一步证实了E钙黏附蛋白、Survivin、Fas对胃癌发生、发展的影响。
Objective To investigate the correlation of E-Cadherin,Survivin and Fas with gastric cancer cell differentiation,and the correlation among E-Cadherin,Survivin and Fas in gastric cancer tissue.Methods Sixty-three cases of well-,moderate-and poor-differentiated gastric cancer tissue and 22 cases of normal gastric mucosa were detected by immunohistochemistry for expression of E-Cadherin,Survivin and Fas.Results The positive rates were 73% for E-Cadherin expression in gastric cancer tissue vs 64% in normal gastric tissue,6% for Fas expression in gastric cancer tissue vs 86% in normal gastric tissue,and 87% for Survivin expression in gastric cancer vs 18% in normal gastric tissue,which reached the level of statistical difference(P0.05).In well-,moderate-and poor-differentiated cancers,the positive rates of E-Cadherin expression were 45%,33% and 5%,respectively,compared with 36%,19% and 10% as found for Fas expression(all P0.05).The trends of positive expression were fairly consistent but not correlated between E-Cadherin and Fas.Although the positive expression of Survivin was gradually enhanced from 68% in well-differentiated cancer to 85% in poor-differentiated cancer,this did not reach the level of significant difference.The expression of Survivin was positively correlated with E-Cadherin(r=0.383,P0.05)and negatively with Fas(r=-0.463,P0.05).However,an overall correlation among E-Cadherin,Survivin and Fas was not found.Conclusion Both E-Cadherin and Fas showed lower rates of positive expression in the gastric cancer tissue than in normal gastric tissue,and were correlated with cell differentiation in a manner contrasted to as found for Survivin.This further proved the role of E-Cadherin,Survivin and Fas in occurrence and progression of gastric cancer,and provided novel theoretical basis for diagnosis and treatment of gastric cancer.
