感染旋毛虫小鼠的心肌CPT1 mRNA表达量及血脂变化 被引量:3

Observation on changes of CPT1 mRNA expression and blood lipid in myocardium of mice infected with Trichinella spiralis

摘要 为探讨旋毛虫感染对宿主脂代谢及心肌损伤的影响,分别取感染前、后不同时期小鼠心肌组织和血清,应用半定量RT-PCR法检测心肌中肉毒碱脂酰转移酶1(CPT1)mRNA的表达量,并对血脂各成分浓度进行测定。结果发现,感染后第14~24天,CPT1mRNA的相对表达量增加明显(P<0.05);由感染前的0.37逐渐增加至0.93(感染后第19天),然后又逐渐下降至0.46(感染后第52天)。表明小鼠心肌CPT1的活性升高,因而心肌细胞不会因滞留过量的游离脂肪酸而造成心肌损伤。血脂中甘油三酯(TG)的浓度均高于对照组的0.43mmol/L,高密度脂蛋白(HDL)的浓度均低于对照组的2.14mmol/L,胆固醇(TC)的浓度下降,低密度脂蛋白的浓度先下降后上升;在感染后第19天,TG上升明显(P<0.05),而HDL、TC下降也比较显著(P<0.05)。可见感染后小鼠脂代谢出现紊乱,可能是导致心肌损伤的重要原因之一。 To explore the influence of Trichinella spiralis infection on host's lipoprotein metabolism and myocardial damage,carnitine palmitoyl-transferase 1(CPT1) mRNA expression was determined using semi-quantitative RT-PCR and blood lipid components were detected during T.spiralis infection in mice.In result,the expression level of CPT1 increased from 0.37 before infection to 0.93 on day 19 post-infection,and then decreased to 0.46 on day 52 post-infection,indicating that bioactivity of CPT1 increased after infection and the metabolism of fatty acids enhanced.Test of plasma lipid showed that the average concentration of TG was higher and HDL was lower compared with that of the control group,0.43 mmol/L and 2.14 mmol/L respectively.These result showed that the mice myocardium suffered from lipids metabolic disorder post-infection with T.spiralis.
