摘要
目的:观察并比较常用不同品系H22肝癌小鼠中医证候的特征与差异。方法:将KM、BALB/c、ICR、C57共4种品系小鼠各30只,分别随机分成正常对照组10只,肝癌组20只。肝癌组小鼠右腋下接种H22细胞造成荷瘤小鼠模型,观察出瘤后各品系肝癌小鼠生存率差异,采用小鼠四诊工作站及其方法,检测小鼠证候的发生与演变。结果:4种品系小鼠腋下接种后均形成实体瘤。在生存率方面,早期各品系肝癌小鼠无明显差异;中晚期差异大,BALB/c肝癌小鼠最低,ICR与KM肝癌小鼠其次,C57肝癌小鼠最高。瘤体积比较,KM肝癌小鼠增长最快,ICR肝癌小鼠增长最慢。去瘤体质量方面,中晚期各荷瘤小鼠呈现下降趋势,ICR小鼠下降最明显,其次是BALB/c与KM小鼠,C57小鼠去瘤体质量变化小。另外,不同品系肝癌小鼠邪毒证与气虚证贯穿疾病始终,且程度逐渐加重,阴虚证与阳虚证在中晚期发生且逐渐加重,其中BALB/c与ICR肝癌小鼠气虚、阴虚、阳虚最严重。结论:KM、BALB/c、ICR、C57等不同品系H22肝癌小鼠均会自发形成证候,KM小鼠最适合于肝癌证候与辨证论治的研究。
Objective: To observe and compare the traditional Chinese medicine syndrome characteristics and difference of H22 tumor mice from different strains.Methods:Thirty mice of KM,BALB/c,ICR and C57 respectively were randomly divided into control group(n=10) and liver cancer group(n=20).The mice in liver cancer group were inoculated with H22 cells to establish the tumor-bearing model and the survival rate of liver cancer mice from different strains were observed.The syndromes of liver cancer mice were detected by four diagnostic method workstation.Results: The solid tumour was formed in liver cancer mice from four strains.The survival rate was no significant difference among four strains mice in early phase;there was significant difference in middle and advanced phase: the survival rate of BALB/c liver cancer mice was the lowest,ICR and KM liver cancer mice followed,and C57 liver cancer mice was the highest.In terms of tumor volume,the tumor in KM liver cancer mice increased fastest and the ICR liver cancer mice increased slowest.The weight of tumor-bearing mice removed tumour were descendent in middle and advanced phase,and ICR mice decreased most obviously,BALB/c and KM mice followed and C57 mice decreased most slowly.The poisonous pathogenic factor syndrome and Qi deficiency syndrome of liver cancer mice from different strains existed in the course of disease and gradually aggravated,yin deficiency syndrome and yang deficiency syndrome occurred in middle and advanced phase and gradually aggravated,and qi deficiency,yin deficiency and yang deficiency were the most serious in BALB/c and ICR liver cancer mice.Conclusion: H22 tumor mice from such different strains as KM,BALB/c,ICR and C57 can form syndromes spontaneously,and KM mice are the optimal model for syndrome differentiation and treatment research.
