摘要
目的筛选气虚证大鼠肾上腺下调的基因。方法采用16周龄Wistar大鼠、自发性高血压大鼠(SHR大鼠)、自发性糖尿病大鼠(GK大鼠),运用标准化大鼠诊法,及Gene Chip Rat Exon1.0 ST Array等技术,检测正常Wistar大鼠、正常气虚Wistar大鼠、SHR气虚和气盛大鼠、GK气虚和气盛大鼠等肾上腺基因表达谱,进而以倍数法筛选3个气虚证肾上腺一致性下调的基因。结果①3个气虚证同时下调的高表达基因有22个,主要包括参与代谢、运输类的Cyp11b3、Hsd3b6、Psmb4、Atpif1、Clic4、Gnai3、Ndufc2、Txnl1;参与信号转导、转录调控类的Jak2、Rhoa、Commd3、Tsc22d1、Polr2g、Capza2。②3个气虚证同时下调中等表达基因有10个,包括参与代谢、运输类的Glyat、Hpd、Slc22a2、Slc7a13;参与转录调控及信号转导类的Pcna、Cbx3、Umod、Serpine1、Emd、MGC108823等。③部分生物学功能不明确的基因,包括Calm2、Papd4、Myd116、Spg21、Emcn、Dhx40、LOC681219、RGD1359616等。结论气虚证Wistar、SHR与GK大鼠肾上腺组织下调表达的基因主要涉及代谢、运输、调控转录、信号转导等生物学过程的紊乱。
Objective To screen and analyse down-regulated genes in adrenal gland of rats about asthenia of 'Qi' syndrome.Methods 16-week-old Wistar rats,spontaneously hypertensive rats(SHR) and spontaneous diabetic rats(GK) were experimented,by the quantitative four diagnosis and syndrome differentiation methods and GeneChip Rat Exon 1.0 ST Array,we observed adrenal gland gene expression about normal Wistar rats,'Qi' asthenia Wistar rats,SHR rats with 'Qi' asthenia and 'Qi' vigorous,GK rats with 'Qi' asthenia and 'Qi' vigorous.Down-regulated genes in three asthenia of 'Qi' syndrome rats were analyzed in this paper.Results(1)Twenty-two down-regulated and high expression genes were gained in three asthenia of 'Qi' syndrome rats.Those genes were involved in metabolism and transport biological process,such as Cyp11b3,Hsd3b6,Psmb4,Atpif1,Clic4,Gnai3,Ndufc2,Txnl1,and involved in signal transduction and transcriptional regulation biological process,such as Jak2,Rhoa,Commd3,Tsc22d1,Polr2g,Capza2.(2)Ten down-regulated and medium expression genes were gained in three asthenia of 'Qi' syndrome rats,it including metabolism and transport biological process genes such as Glyat,Hpd,Slc22a2,Slc7a13,and including transcription regulation and signal transduction genes such as Pcna,Cbx3,Umod,Serpine1,Emd,MGC108823 and so on.(3)Some genes biological function was not clear,for example,Calm2,Papd4,Myd116,Spg21,Emcn,Dhx40,LOC681219,RGD1359616 and so on.Conclusion Down-regulated genes about 'Qi' asthenia Wistar,SHR and GK rats adrenal gland are mainly related to the disorder of biological processes such as metabolism,transport,regulation of transcription,signal transduction.
