摘要
近年来,以焦虑、抑郁等负性情绪为主的经前期综合征(PMS)等中医典型情志病证正呈高发态势,但其病理生理学机制仍模糊不清。PMS/经前烦躁症(PMDD)病证结合动物模型可以模拟人类PMS/PMDD的临床症状并进行症状的药物逆转研究,对于PMS/PMDD发病机制及治疗药物研发、临床疗效评价都有着重要的应用价值。文章纵览近年相关研究文献,综述PMS/PMDD自发性动物模型和诱发性动物模型并指出环境应激、社会应激和神经生化等3类诱发性动物造模方式。而动物模型是否可靠需再经过动物模型表面效度、结构效度及预测效度进行综合评价,进而形成成熟的实验动物模型,满足研究所需。实验动物种类、评价指标与造模方法相辅相成,有机结合构建出PMS/PMDD动物水平研究基石。
In recent years,TCM syndromes such as premenstrual syndrome,which are mainly negative emotions such as anxiety and depression,are showing high incidence,but their pathophysiological mechanisms are still unclear.Aimal models combined with PMS/PMDD and syndrome can simulate the clinical symptoms of human PMS/PMDD and conduct drug reversal studies of symptoms.It has important application value for the pathogenesis of PMS/PMDD,therapeutic drug development and clinical efficacy evaluation.In this paper,we review the relevant research literatures in recent years,review the PMS/PMDD spontaneous animal models and induced animal models and point out three types of induced animal modeling methods:environmental stress,social stress and neurobiochemistry.Whether the animal model is reliable or not needs to be comprehensively evaluated by the animal model’s face validity,construct validity and predictive validity,and then form a mature experimental animal model to meet the research needs.The experimental animal species,evaluation indicators and modeling methods complement each other,and organically combined to construct the cornerstone of PMS/PMDD animal level research.
