急性心肌梗死动物模型应用分析

Application Analysis of Animal Models of Acute Myocardial Infarction

摘要 目的 研究急性心肌梗死(acute myocardial infarction,AMI)动物模型复制要素,提供已有急性心肌梗死模型的应用现状及特点,为相关研究提供模型的选择与参考。方法 以“急性心肌梗死”“动物”为主题词,在中国期刊全文数据库(CNKI)、万方数据库(Wanfang)、维普数据库(VIP)和PubMed等平台进行检索,时间从2017年1月至2021年12月,收集实验动物种类、性别、模型复制方法、检测指标、麻醉方法、给药周期等,建立数据库,使用Excel 2013对收集的数据进行统计分析。结果 纳入符合标准的期刊文献142篇,其中使用最多的实验动物种类为SD大鼠,常选择结扎冠状动脉的方式复制急性心肌梗死动物模型,以腹腔注射戊巴比妥钠的方式对实验动物进行麻醉,以心电图病理性改变为模型复制成功标准,对心功能、血流动力学等生物机能指标进行检测,多采用先模型复制后给药的实验方式,给药周期为28 d。结论 急性心肌梗死模型复制方法比较成熟,成模率高、可复制性强,但是现有检测标准中缺少表观指标的观察,研究药物机制多从单一因素入手,未来应增加此类研究,为临床新药开发和进一步药物机制研究提供可靠依据。 Objective To study the modeling elements for animal models of acute myocardial infarction(AMI),to provide the application and characteristics of existing AMI models,and to provide model selection and reference for related studies. Methods Using "acute myocardial infarction" and "animal" as keywords,we searched the CNKI,Wanfang, VIP and PubMed from January 2017 to December 2021, and collected experimental animal species,sex,modeling method,testing index,anesthesia method,and drug administration cycle. After that we established a database and used Excel 2013 for statistical analysis of the collected data. Results A total of 142 articles meeting the criteria were included. It was found that SD rats are the most used experimental animal species. The acute myocardial infarction model was replicated by ligating the coronary artery. The experimental animals were anesthetized by intraperitoneal injection of sodium pentobarbital. Pathological changes in the electrocardiogram are a measure of successful reproduction of model. Cardiac function, hemodynamics, and other biological function indexes were tested. The experiment was conducted by modeling at first and then drug administration,and the drug administration period was 28 days. Conclusion The method for modeling AMI is relatively mature, with high modeling rate and strong reproducibility. However,there is a lack of observation of apparent index in the existing detection standards,and the study of drug mechanisms mostly starts from a single factor. These findings suggest that related studies should be increased in future to provide a reliable basis for new drug development and further research of drug mechanism.
