摘要
目的:从不同根系土壤分离筛选高效降解有机磷的细菌.方法:通过平板法进行初筛,根据水解圈直径和菌落直径比值大小筛选到水解圈直径和菌落直径比值较大的菌株,进一步通过液体摇瓶培养复筛,对其解磷能力进行了测定,并通过形态特征、生理生化特性和16S rDNA序列测定等一系列试验对其进行鉴定.结果:分离纯化出10株解磷菌,经平板法初筛出6株水解圈直径和菌落直径比值较大的菌株,最后复筛出1株分解有机磷能力较强的菌株XK-1,解磷能力达22.072mg/L.该菌株鉴定为枯草芽孢杆菌.结论:成功分离出一株解有机磷能力较强的枯草芽孢杆菌XK-1.
Objective:to isolate and screen organ ophosphate-solubilizing bacterium from the different rhizos-phere and soil.Method:through the plate method for the first screening,we get bacterium with the larger ratio of hydrolysis circle diameter and colony diameter.Further screening in shake culture experiment,it was identified by a series of bacterial colony morphological,physiological characteristics and 16 S rDNA sequences and other tests.Results:of these strains,we get one the better strains XK-1 in organic ophosphate-degradation ability to 22.072 mg/L.It are identified as Bacillus subtilis.Conclusion:Bacillus subtili XK-1 is screened successfully which can excrete high ability of dissolving organic phosphate.
