摘要
JM-35菌株是从鸡肠道中分离得到的一株能够抑制致病性大肠杆菌(E.coli)的益生菌,为提高该菌株的芽孢生成率,以便于今后工业化生产,对其产芽孢条件进行了优化。采用单因素试验和正交试验方法,优化出JM-35菌株的最适宜的发酵培养基组成为:麦麸3%,胰蛋白胨1%,MnSO4.H2O 0.05%,CaCl2.2H2O0.1%,并且进一步确定发酵产芽孢条件:发酵时间72h,装瓶量50mL/250mL,接种量2%,初始pH值6.0。最终通过优化后的培养基进行发酵,JM-35菌株发酵液中的芽孢生成率提高到95.70%,细菌总数达到1.95×109 cfu/mL。
Research was done to improve ratio of spore production of Probiotics JM-35 from fowls, which could strongly inhibit Escherichia coli, the main pathogens causing fowls disea- ses. By single factor and orthogonal experiments, the most suitable carbon source, nitrogen source and inorganic salts to spore production of strain JM-35 were chosen. With range analy- sis, the most suitable fermentation medium and spore production conditions of strain JM-35 were determined as follows: media composed of bran 3. 0%, tryptone 1.0%, MnSO4 · H2O 0.05 %, CaCl2 · 2H2 O 0.1%, culture time 72 h, bottle filling capacity 50 mL/250 mL ,inoculum size 2%and pH 6.0. Under this optimum condition, the rate of spore production was 95.70% and the total bacterial of strain JM-35 reached 1.95 × 109cfu/mL .
