摘要
建立了柱前衍生化-HPLC法同时测定三七中的18种游离氨基酸。以异硫氰酸苯酯为柱前衍生化试剂,采用C_(18)色谱柱,以乙腈-0.05 mol/L乙酸钠缓冲液为流动相,梯度洗脱,检测波长254 nm。结果显示三七根中的游离氨基酸以精氨酸、丙氨酸和脯氨酸为主,约占18种游离氨基酸总量的80%。18种氨基酸的平均回收率为96.5%~103.4%,RSD均小于3%。
A pre-column derivatization HPLC method was established for the determination of 18 free amino acids in Panax notoginsens. Phenylisothiocyanate was used as a derivatizing agent. A C18 column was used with the mobile phase of acetonitrile-0.05 mol/L sodium acetate buffer, at the detection wavelength of 254 nm. The results showed that the major components were arginine, alanine and proline, accounting for approximate 80% of free amino acids. The average recoveries were 96.5 %-103.4 %, with RSDs less than 3 %.
