摘要
目的建立哈药别克参(新疆猪牙花肉质鳞茎)的氨基酸成分的TLC定性及HPLC法定量鉴别。方法 TLC法以正丁醇-冰醋酸-水(4∶1∶1)为展开剂和0.2%茚三酮乙醇试剂为显色剂。HPLC法采用ACQ柱前衍生,Hypersil BDS C18色谱柱(5μm,4.6 mm×250 mm),流动相A为乙腈-醋酸钠缓冲盐溶液(7∶93,pH=6.5),流动相B为甲醇-乙腈-水(20∶60∶20),线性梯度洗脱。结果 TLC法鉴别出别克参中所含精氨酸、缬氨酸、苯丙氨酸和苏氨酸4种氨基酸,斑点清晰,分离度高。HPLC法测定别克参中13种氨基酸。结论别克参的氨基酸成分以精氨酸、苏氨酸和谷氨酸的含有量为多。
AIM To establish the identification and quantitative method for analyzing amino acids from Eryth- ronium sibiricum's bulb by TLC and HPLC. METHODS A TLC method was developed with n-butanol-acetic acid-water (4 : 1 : 1 ) as the developer, and 0. 2% ninhydrin ethanol reagent as chromogenic reagent. A pre-col- umn derivatization HPLC method was used for the measure of the contents of amino acids. The separation was car- ried out on a Hypersil BDS Cls column (5 μm, 4.6 mm ×250 mm) in a gradient elution mode. The mobile phase was consisted of A ( acetonitrile-sodium acetate solution, 7 : 93, pH = 6.5 ), and B ( methanol-acetonitrile-water, 20 : 60 : 20). RESULTS TLC analysis indicated that the test samples contained arginine, valine, phenylalanine and threonine. The spots were clear and the separation was high. HPLC analysis revealed that the test samples had 13 kinds of amino acids. CONCLUSION The contents of arginine, threonine and glutamic acid are higher in Erythronium sibiricum' s bulb.
