鞘内注射氟代柠檬酸对切口痛大鼠痛阈和脊髓TNF-α mRNA表达的影响 被引量:2

Effect of Intrathecal Injection of Fluorocitrate on Pain Threshold and Expression of TNF-α mRNA in Spinal Cord of Rats with Incision Pain

摘要 目的观察鞘内注射氟代柠檬酸对切口痛大鼠痛阈及脊髓组织TNF-α mRNA表达的影响。方法取60只雌性SD大鼠制备切口疼痛模型,并随机分为对照组和实验组,每组30只,手术后分别鞘内注射10μL生理盐水、1 nmol氟代柠檬酸(10μL)。测量两组大鼠手术前、手术后4、8、16及24 h机械痛阈、热痛阈及脊髓组织TNF-α mRNA表达。结果手术后,大鼠机械痛阈和热痛阈均较术前下降(P<0.05或P<0.01),且术后实验组均高于同一检测时点的对照组(P<0.05或P<0.01);脊髓组织TNF-α mRNA表达上调(P<0.05或P<0.01),实验组低于对照组(P<0.05或P<0.01)。结论脊髓胶质细胞激活并促进TNF-α mRNA表达上调可能参与了切口痛的发生和维持。 Objective To observe the effect of intrathecal injection of fluorocitrate on pain thresholds and the expression of TNF-α mRNA in spinal cord of rats with incision pain. Methods Sixty female Sprague-Dawley rats were randomly divided into two groups (n= 30 each) :control group receiving intrathecal injection of 10μL physiological saline after a 1-cm longitudinal incision was made through skin, fascia and muscle of the plantar aspect of the left hindpaw;test group receiving intrathecal injection of 1 nmol fluorocitrate(10μL)after operation. Mechanical and thermal pain thresholds were measured by Dynamic Plantar Aes thesiometer( UGO BASILE, Italy) and Plantar test 7375 ( UGO BASILE, Italy) before and 4,8,16,24 h after operation. The expression o( TNF-α mRNA in spinal cord before and 4,8,16,24 h after operation was detected by RT-PCR. Results Mechanical and thermal pain thresholds were decreased after operation in two groups(P〈0.05 or P〈0.01), and those in the test group were higher than those in the control group(P〈0.05 or P〈0. 01). The expression of TNF-α mRNA in spinal cord was increased after operation in two groups(P〈0.05 or P〈0.01) ,and that in the test group was lower than that in the control group (P〈0. 05 or P〈0.01). Conclusion The activation of gliacyte in the spinal cord may be involved in the modulation of incision pain in rats.
